Semax (Met-Glu-His-Phe-Pro-Gly-Pro) is a synthetic heptapeptide based on the ACTH(4-7) fragment, developed at the Institute of Molecular Genetics of the Russian Academy of Sciences. It has been approved in Russia and several CIS countries since 2011 for treatment of stroke, cognitive disorders, and as a nootropic (cognitive enhancer).
Unlike the full ACTH molecule, Semax lacks hormonal (adrenocorticotropic) activity — it does not stimulate cortisol production. Instead, it acts as a neurotrophin modulator, increasing brain-derived neurotrophic factor (BDNF), nerve growth factor (NGF), and other neurotrophins that support neuronal health, plasticity, and repair.
Semax is administered intranasally for rapid brain delivery and has been extensively studied in Russian clinical practice for over two decades. It represents a well-characterized nootropic with an established clinical track record, even if most evidence is published in Russian-language journals.
