// Overview

What is TB-500?

TB-500 is a synthetic peptide representing the active region (amino acids 17-23) of Thymosin Beta-4 (Tβ4), a naturally occurring 43-amino acid protein found in nearly all human cells. Thymosin Beta-4 is the primary intracellular G-actin sequestering protein, playing a fundamental role in cell motility, migration, and tissue repair.

While the full Thymosin Beta-4 protein is large and expensive to synthesize, the TB-500 fragment retains the key actin-binding domain responsible for its therapeutic effects. This fragment promotes cell migration to injury sites, stimulates new blood vessel formation (angiogenesis), reduces inflammation, and supports the formation of new muscle and skin tissue.

TB-500 is often paired with BPC-157 in what the peptide community calls the “healing stack” — BPC-157 drives growth factor signaling while TB-500 handles cellular migration and tissue remodeling, creating complementary healing pathways.

// How It Works

How TB-500 Works

TB-500 promotes healing through actin-mediated cellular repair:

  1. Actin binding and cell migration — The LKKTETQ sequence binds G-actin, promoting actin polymerization which drives cell motility. This allows immune cells, fibroblasts, and stem cells to migrate to injury sites faster.
  2. Angiogenesis — Stimulates formation of new blood vessels from existing vasculature, improving blood supply to damaged tissues. This is critical for healing avascular or poorly vascularized injuries.
  3. Anti-inflammatory action — Downregulates inflammatory cytokines and chemokines, reducing the inflammatory phase and allowing earlier transition to the repair/remodeling phase.
  4. Matrix metalloproteinase regulation — Modulates MMP activity to support tissue remodeling without excessive scarring or fibrosis.
  5. Stem cell activation — Promotes differentiation and migration of resident stem cells, particularly cardiac progenitor cells and keratinocyte stem cells.

Level Dose Frequency Duration
Loading 2-2.5 mg 2x per week 4-6 weeks
Maintenance 2 mg 1-2x per week 4-8 weeks
Acute Injury 2.5 mg 2x per week 2-4 weeks
Systemic 5 mg 1x per week 8-12 weeks

What to Expect

Week 1-2

Reduced inflammation at injury sites. Improved flexibility and reduced stiffness. Some users notice better hair/nail growth early.

Week 3-4

Healing of chronic injuries accelerating. Range of motion improving. Skin healing faster. Workout recovery noticeably better.

Week 5-8

Significant tissue repair. Chronic tendon/joint issues resolving. Hair density may improve. Scars fading.

Week 8-12

Continued remodeling of healed tissue. Best results when combined with BPC-157 for the full healing stack.
